Ich hatte bei mir mal vacation getestet mit:

#if header :contains ["subject"] ["vacationtest"] {
# vacation :days 1 :addresses ["invalid at japantest.homelinux.com"] :subject 
"On vacation messages" "bin wegweg";}

Bei dir fehlt anscheinend der Auslöser. vacation ist eine Aktion, aber du 
hast keine Bedingung für die Aktion angegeben.

Beachte bitte, dass vacation eine Mail nur einmal losschickt.
